What kind of automated Elispot reader system is used for the plate evaluation service?

We are using the high resolution automated Elispot reader system from Carl Zeiss with its newest KS Elispot software version.

What does high resolution mean?

There are in general 2 different reader systems out there. Most reader systems take one picture per well. The resolution of the picture taken depends therefore only on the camera used, and can be expressed in the pixel size (smallest unit of a picture). With one picture per well, the average pixel size is 15 - 35 µm. This should be referred to as low resolution in Elispot evaluation. With a high resolution system, multiple pictures per well are taken. The resolution is therefore not only dependent on the camera, but also on the magnifying system used (microscope). The average pixel size is 2 µm,and the resolution is multiple times higher than in a low resolution system.

Why high resolution

Why is high resolution important for the evaluation of Elispot plates?

An accurate image analysis requires at least 7 pixels (smallest unit of a picture) across the object to be analyzed. The average spot size (= object) is 20 -100 µm in diameter. With a high resolution reader (pixel size = 2 µm), this translates into 10 - 50 pixels across the spot. An accurate analysis of a spot including the recognition and exclusion of artifacts is possible. A low resolution system (pixel size in average 15 µm) can only provide 1 - 7 pixels across the spot. Small spots and artifacts appear as rectangles and look very much alike. No accurate evaluation can be assured.

Can a reader perform a high resolution evaluation if it takes only 1 picture per well?

The resolution of readers taking one picture per well depends on the camera, especially their chip size which determines the average pixel size of the picture of one well of a 96 well plate. The most sophisticated analog cameras are able to reach a pixel size of 9 µm. Most readers use cameras offering a pixel size of 15 - 35 µm. Keeping in mind that 7 pixels are needed across the diameter of a spot for accurate evaluation, only spots larger than 7 x 9 µm = 63 µm are evaluated accurately. High end digital cameras are able to slightly decrease the pixel size. However, they increase the evaluation time and are very expensive. For Elispot evaluation purposes, pixel sizes reached with systems taking one picture per well should be referred to as low resolution systems. Objects (spots, artifacts) below 60 µm in diameter can't be analyzed precisely.

Can ZellNet evaluate double stained plates?

Yes. The KS Elispot software allows us to adapt the evaluation parameters for specific spot features including the color range. Double stained plates are evaluated for 3 color ranges (typically red, blue and purple for double stained spots).

Can fluorescent Elispot samples be evaluated with the Zeiss reader?

Yes. The reader system is able to evaluate fluorescent Elispot samples as well.

Scanning and Evaluation

Is there a difference between scanning and evaluation service?

In general, yes. Scanning services do not include the actual evaluation of your wells. ZellNet offers an evaluation service that includes scanning by default. This means, that all wells are scanned and evaluated at the same time. There is no additional time required for evaluation after scanning the wells. However, scanned pictures can be stored and re-evaluated, if needed.

Is there any specific software required on the customer end for analyzing the pictures scanned by ZellNet?

No, there is no software requirement. ZellNet's service includes scanning and evaluation. You will get multiple data and picture files from the evaluation of your plate.

Do we need to use certain Elispot plates to be able to have them evaluated by ZellNet?

No, there is no plate requirement. Any type of plates used for Elispot assays can be evaluated by us.

Yes, we are able to evaluate clear plastic plates. There is no restriction on the plates which can be sent for evaluation.
