I am wondering if you found an answer to your query. I too am doing murine elispot and having the exact same problem. Immunized mouse spleen cells, rbc depleted, in 10% heat inactivated FBS. IL-17 no problem, but IFN-g has given me real issue. They are very "REAL" spots so I am wondering about the processing measures taken to rbc deplete. I have used IFN-g from Mabtech, R+D, BD Pharm and I get the same issue with all of them. ANy ideas??
