Sylvia, Thanks for your suggestions with this, we are currently using the nitrocellulose HA plates from millipore, do you find that the IP plates work better? Another question you may be able to shed some light on... we add the peptides directly to the splenocytes(5 x 10^5 cells/well) relying upon APCs within the splenocyte prep. Do you recommend adding the splenocytes to the wells, incubating an hour then carefully adding the peptides or adding the peptides first (as was suggested by another group performing the same procedure) then the splenocytes or does it make any difference at all? Would appreciate any advice, thanks again. Kim
