Subject Coating with peptide

I am new to the elispot assay and am trying to use the technique to identify influenza peptide-specific AFC. We would like to coat the plates with the peptide instead of a primary antibody, and then use isotype-specfic antibodies to identify different AFC populations. Is there any reason to think that coating plates with peptide would be any different than with antibody? Should I stick with the 10 ug/ml concentration as recommended in the Millipore protocol for the IFNg elispot?
