I have been trying to develop a protocol for doing mouse ELiSPOTs, specifically to detect mouse IL-2 and mouse IFN-g. So far, I have had absolutely NO LUCK. I am using antibodies created for doing ELISAs from PharMingen and have attempted to titrate them. The problem is that my negative and positive (ie "non-immunized" and "immunized" mouse spleen stimulated with Con A) samples look identical (lots of REAL looking spots). All of my incubations and washes are the same as a neighboring lab whose human ELiSPOTS seem to work beautifully. I am suspecting the problem is in the preparation of the cells. Could the problem be using the whole spleen or in the stimulation (I know in human they use purified cell populations)? Has ANYONE out there done a mouse ELiSPOT that works??!?! Please help!!! Thanks.
