Hi Cathy,
I have been trying to find plate washer conditions that would work as well as the squeeze bottle, but no luck yet. What I've learned is that the more volume you use, the more spots you get, unfortunately the spots in the center are more intense than the rest of the membrane. The squeeze bottle is time consuming and in addition it can really make your hand sore when you are doing a lot of plates (20+). What we've done in our lab is put a 20L jug on a high shelf, connected it to some tubing and then cut the end of the sgueeze bottle off and stuck in on the end of the tubing. It works great! It still takes the same amount of time to wash the plates but your hand feels much better. I wouldn't use a plate washer in ELISPOT development. I hope that information was useful.
