Hi! In my experience, it takes a while to optimize the protocol for
your needs. Blocking with PBS/BSA should be fine. However, you
can also use your RPMI + serum. If you get spots without adding cells
(did I understand this right?) then this might be due to whatever you
use for coating. Spots in wells without cells can also be due to
the FCS you are using or some infection in development buffers.
Which method do you use? Avidine likes to bind to many things,
also bacterial proteins from special strains - this can cause
"beautiful" spots.
