It is not unusual to see differences between the number of GrB and IFN-g spots. One reason is that not all CTL that secrete GrB also secrete IFN-g. It could also be a function of your stimulant. We have found that different peptides can elicit differnt types and manitudes of response. Additionally, it seems that peptides with high binding avidity/affinity elicit a better GrB response.
